I've been gathering some recent info about weapon and car customization from the web;
Weapon customization:

There will be a wide range of weapon modifications, here's a few examples;
- Extended clips (more ammo)
- Grips (improve accuracy)
- Weapon tints (custom colors)
- Flashlights
- Scopes
- Surpressors (which will allow you to complete some missions in stealth mode)

There will be a 'locomotion system' that will adapt your movement and behaviour depending on if you're just walking around, or firing/in battle. In the latter case you'll change over to a 'combat jog', and the way you handle and move your weapons will even be depending on which of the 3 characters you're playing at that moment.
You'll be able to choose from 3 firing modes;
Free aim,
assisted aim (slightly larger targeting area and threat indication) and
traditional GTA (comparable to GTA IV's soft lock)
Car Customization:
Vehicle customization is back (it was possible in San Andreas too), and this time more extensive then ever before, with not only cosmetic adaptations, but also customization that will affect your gameplay. This time there won't be "Pay and Spray's", but you'll have several one stop "LS Custom's" dotted around the landscape that will be able to handle your customization and repairs;

All in all there will be over 1,000 car (and bike!) modifications available in the game, so looks like there's more then enough choice to have some fun with this.
Personally I really like car modification in open world games like GTA, not just because of looks and functionality, but I especially like how it makes you take care of 'your' car, and I always find myself putting more effort into preventing damaging it, and doing frequent repairs instead of just discarding your wreck and stealing the next anonymous car.
Can't wait for this game! Luckily we can start counting down in weeks instead of months now
